Penn Station is in the greater nyc area. Manhattan is the main borough of NYC, where most of the main attractions and where all the tourists go. Penn Station is right in the heart of midtown manhattan, close to many of the main attractions. Madison Square Garden is right upstairs, Macy's is a block away, Empire State Building is a couple blocks away, Times Square is 10 blocks away. There is a big shopping area right near Penn Station as well.

Penn Station will land you right in the middle of Manhattan, then you just need to get in a taxi, on the subway, or simply walk to where you want to go.
